Understanding the molecular signatures of endometrial receptivity and ovulation for the contraception development and therapeutics exploration for endocrine and metabolic gynecological disorders, PCOS and Endometriosis

Population stabilization is essential to meet a healthy life and socio-economic status. In India, the population is one of the major challenges to meet the optimal resources. We are working on contraceptive agents for women using various modern biology tools. We have explored various endometrial signal transduction pathways at kinase, Small G-protein, epigenetics, etc. level, which is essential for its receptivity for blastocyst implantation to establish the pregnancy. We found Integrin beta8 mediated TGF-β/Endoglin and RAC1 activation in the endometrial receptivity (Biology of Reproduction 2013 and 2015, Molecular Cellular Endocrinology 2016, and Scientific Reports 2017). Further, epigenetically miR-149 also regulates PARP-2, an essential molecule for endometrial receptivity and decidualization along with PARP-1 (Reproduction 2014, Molecular and Cellular Endocrinology 2020, Molecular Human Reproduction 2021 and Reproductive Sciences). Apart from the population instability, lifestyle pattern also gives rise to, endocrine and metabolic disturbance, leading to various disorders; Polycystic ovarian syndrome (PCOS) and endometriosis. These disorders' occurrence is more than 10% among the reproductive age women and induces anomaly at reproductive as well as other health issues. We are trying to understand the mechanism of PCOS and endometriosis etiologies using modern biology tools using rodent models and in vitro models. We also collaborate with a gynecologist to look at the clinical aspect as well as to remains in touch with the prevailing situation of the disorders. Further, we have models in place to screen the compounds (synthetic and natural resources) and compounds-specific fractions for the therapeutic potential of the above disorders.

Some of our research group broad goals includes,

• Elucidating the molecular mechanism of the endometrial receptivity for the blastocyst implantation failure associated with endometriosis and polycystic ovary syndrome pathophysiology.

• To examine the endocrine disturbance driven alteration of in the intraovarian factors (PCOS) and endometriotic lesion development (Endometriosis).

• Exploration of management of the Polycystic ovarian syndrome (PCOS) and Endometriosis gynecological disorders using rodent models.
